Scarlet Spider is a perfected synthezoid created by Doctor Octopus and Arnim Zola using Spider-Man's DNA. Initially, Scarlet Spider had no knowledge of his true origins, as Doctor Octopus had implanted false memories into his brain and told lies to the former in order to retain his loyalty. This led Scarlet Spider to believe he was born as a normal human and grew up as a street orphan before Doctor Octopus "saved" him and gave him spider-like powers at the cost of his memories.

As part of Doctor Octopus's plans to defeat Spider-Man, Scarlet Spider is sent into S.H.I.E.L.D. as a spy and joins the Web-Warriors. During his time with the superheroes, he meets May Parker and is given the name "Ben Reilly", after May's late husband, Ben Parker. Touched by the kindness and respect Spider-Man and his aunt has shown him, he betrays Doctor Octopus and presumably sacrifices his life to prevent Doctor Octopus's Octopus Island from destroying New York City. Scarlet Spider survives the crash, but he keeps this a secret and refuses to return to the Web-Warriors out of shame for his past actions. He decided to search for answers about his identity, which eventually leads him to reunite with Spider-Man. After discovering the truth of his origins, Scarlet Spider becomes infuriated and disillusioned, but with encouragement from Spider-Man, he overcomes his initial programming and resolves not to let his origins affect him. Now focused on establishing his own identity as an independent being, Scarlet Spider rejoins the Web-Warriors as a true member of the team.

• Scarlet Spider's incarnation in the Ultimate Spider-Man series is an amalgam of the first Scarlet Spider, Ben Reilly, and the fourth and current Scarlet Spider, Kaine. In The Amazing Spider-Man comic book series, Ben Reilly and Kaine are both clones of Peter Parker created by Jackal. While this incarnation of Scarlet Spider shares Kaine's costume, abilities (e.g. his stringers and organic webbing), and belligerent personality, he takes on the name "Ben Reilly" in the series. However, unlike in the comic book series, Scarlet Spider is a synthezoid/human hybrid created by Doctor Octopus and Arnim Zola.
