Curtis "Curt" Connors is a S.H.I.E.L.D. scientist. After Spider-Man is recruited into the agency, Connors quickly befriends the young superhero, who views him as trustworthy friend and colleague. When the Green Goblin attacked the Helicarrier, Connors's right arm is crushed by falling debris, forcing him to have it amputated. While investigating Doctor Octopus's various animal DNA formulas, Connors realizes the potential medical benefits they could provide to the world, and injects himself with lizard DNA in hopes of restoring his right arm. Although his arm is successfully regrown, the serum causes Connors to transform into a reptilian humanoid monster known as the "Lizard".
